Legionella: Molecular Microbiology
نویسنده
چکیده
The fi rst 3 chapters of Legionella: Molecular Microbiology deal with clinical aspects of legionellosis. Paul Edelstein provides a fascinating glimpse into the history of Legionnaires’ disease, revealing little-known facts from the perspective of one who participated in the investigation of the 1979 outbreak at Wadsworth Veterans Administration Hospital in Los Angeles, California. Although the chapter on epidemiology focuses on Legionnaires’ disease in Europe, it provides new information from the European Working Group for Legionella Infections. The remaining 9 chapters cover various aspects of the molecular biology of Legionella. These chapters provide up-to-date information for basic scientists, but they are also of value to clinicians. However, the chapter on genetics and immunology of host resistance to Legionella infections is illuminating, yet disappointing—illuminating because it shows that much has been learned but disappointing in that we still do not understand much about the biology of Legionella infections. It is also disappointing that the efforts of clinicians and basic scientists are not coordinated at the time of outbreaks, so material could be collected that would enable more in-depth study of the genetics and immunology of host resistance. The book provides a glimpse into the challenges of studying an intracellular pathogen that is found in human macrophages and in amebae. For anyone who studies intracellular pathogens, the chapter on mechanisms of intracellular survival and replication of L. pneumophila will be instructive. One minor distracting feature is that the introduction to most chapters tends to repeat the same material about Legionnaires’ disease. Also, a background in molecular biology would be helpful in understanding some of the more technical chapters, although it is not absolutely necessary. Overall, not only will this book be pertinent to all who study Legionella spp. and other intracellular microorganisms in the laboratory, but it will also be a valuable reference for infectious disease clinicians, microbiologists, and public health professionals.

متن کاملMulticenter comparison of molecular methods for detection of Legionella spp. in sputum samples.
Legionellosis can be diagnosed by PCR using sputum samples. In this report, the methods of nine laboratories for 12 sputum samples with Legionella pneumophila and Legionella longbeachae are compared. We conclude that (i) liquefaction prevents PCR inhibition, (ii) the employed mip gene PCRs detected L. pneumophila only, and (iii) the 16S rRNA gene PCR detected both Legionella species and is pref...
متن کاملLegionella micdadei, a new cause of prosthetic joint infection.
Extrapulmonary infections caused by Legionella spp. other than Legionella pneumophila are rare. We report what is, to our knowledge, the first description of a prosthetic joint infection due to Legionella spp. Systematic testing of samples with suspected prosthetic infection by molecular biology techniques was essential. Legionella micdadei should be added to the list of microorganisms causing ...
متن کاملIntracellular parasitism and molecular determinants of Legionella virulence.
Bacteria of the genus Legionella are intracellular parasites and major human pathogens. They bind to surface receptors, penetrate eukaryotic cells and initiate complex disorders during phagocytosis. These disorders include inhibition of oxidative burst, a decrease in phagosome acidification, the blocking of phagosome maturation and changes in organelle trafficking. As a result, the microorganis...
متن کاملDetermination of catalase, peroxidase, and superoxide dismutase within the genus Legionella.
We examined 40 strains of Legionella for reduced-oxygen scavenging enzymes. Using a simple reaction chamber with a Swinney filter for the Beers and Sizer assay, we determined the catalase activity of live cells grown on buffered charcoal-yeast extract agar. For 29 strains of Legionella pneumophila, the apparent first-order rate constants for catalase ranged from 0.000 to 0.005. Similarly, low v...
متن کاملPersistence of Legionella in hospital water supplies and nosocomial Legionnaires' disease.
The molecular epidemiology of clinical and environmental Legionella species isolates was studied in seven hospitals from 1989 to 2006. The number of environmental pulsed field gel electrophoresis (PFGE) patterns ranged from one to nine according to the hospital. Genomic PFGE pattern persistence was observed in 71% of the hospitals, even after 17 years in some hospitals, and the relationship bet...
